Self-proclaimed “background character” Kazuhiko Nukumizu accidentally witnesses Anna Yanami, a popular girl in his class, being rejected by her childhood friend. Nukumizu later continues getting roped in to helping “losing heroines” – mostly characters embodying archetypes of the childhood friend romance trope – into resolving their romantic conflicts.

Nukumizu, Anna, Lemon, and friends are students of a school modeled after Aichi Prefectural Jishukan High School in Toyohashi, Aichi prefecture. Makeine is an outlier in terms of how localized its spots are, at least for the material covered in the Summer 2024 season of the anime. Every location depicted is either in downtown Toyohashi or its neighboring region. This makes it prime as a tool for contents tourism promotion. The series has collaborated with Toyohashi Zoo and JR Tokai. In addition, standees and official prints have been displayed in locations like Mokkuru Shinshiro Michi-no-eki, Coffee And Canele, Bon Senga, Seibunkan, Tokiwa Shopping Street, the Toyohashi City Hall, and Toyohashi Suijo Building Mall.
